
This product is intended for the in vitro quantitative determination of tacrolimus, cyclosporine, and sirolimus concentrations in human whole blood for use as an aid in clinical diagnosis.
Tacrolimus is indicated for the prevention of organ rejection in liver and kidney transplant recipients. It exerts its immunosuppressive effect by binding to the specific FK506-binding protein (FKBP) in T lymphocytes, thereby inhibiting the gene transcription and protein expression of cytokines associated with T-cell activation. Cyclosporine (also known as Cyclosporin A) is also a calcineurin inhibitor with potent immunosuppressive activity. It is widely used to prevent post-transplant immune rejection and is primarily indicated for the prevention and treatment of graft-versus-host disease (GVHD) following bone marrow transplantation. Sirolimus is mainly used in liver transplant recipients with impaired renal function, intolerance to calcineurin inhibitor (CNI)-related adverse effects, or an increased risk of tumor recurrence.
Because these immunosuppressive agents have narrow therapeutic windows and exhibit significant intra- and inter-individual variability, therapeutic drug monitoring (TDM) is mandatory in clinical practice for tacrolimus, cyclosporine, and sirolimus.
