
This assay is intended for the in vitro quantitative determination of α1-antitrypsin (AAT) in human serum or plasma as an aid in clinical diagnosis.
Serum AAT measurement is widely used in the evaluation of inflammatory conditions, liver disease, and protein-losing disorders.
Elevated AAT concentrations are commonly observed in acute and chronic inflammatory conditions, including infections and rheumatoid diseases, as well as in trauma and tissue necrosis. Decreased AAT levels are frequently associated with severe liver injury, nephrotic syndrome, malnutrition, and other conditions affecting AAT synthesis or excessive protein loss.
α1-Antitrypsin is a 54 kDa glycoprotein synthesized primarily by the liver. As the major serine protease inhibitor in human serum, it inhibits a variety of proteolytic enzymes, including neutrophil elastase, thrombin, urokinase, and other proteases. AAT also functions as an acute-phase protein. During inflammatory responses, increased vascular permeability allows AAT to diffuse into the interstitial fluid, where elevated local concentrations help limit protease-mediated tissue injury and contribute to the regulation of acute inflammation.
α1-Antitrypsin present in the sample specifically reacts with polyclonal anti-human α1-antitrypsin antibodies contained in the reagent to form insoluble immune complexes, resulting in increased turbidity. The degree of turbidity is directly proportional to the α1-antitrypsin concentration in the sample.
By measuring the turbidity at 340 nm and applying a multipoint calibration curve, the α1-antitrypsin concentration in the serum or plasma specimen can be accurately quantified.
