  • Clinical Background

    This assay is intended for the in vitro quantitative determination of haptoglobin (HPT) in human serum or plasma as an aid in clinical diagnosis.

    Haptoglobin is primarily used as an important biomarker for the assessment of intravascular hemolytic disorders.

    Also known as hemoglobin-binding protein, haptoglobin is an acute-phase glycoprotein synthesized by hepatocytes. It consists of two α (alpha) chains and two β (beta) chains. Haptoglobin binds free hemoglobin released during hemolysis and transports the resulting complex to the liver for degradation.

    During intravascular hemolysis, haptoglobin is consumed much more rapidly than it can be synthesized by the liver. Consequently, any condition associated with increased erythrocyte destruction and elevated free hemoglobin leads to a reduction in serum haptoglobin levels. Serum haptoglobin concentrations are markedly decreased in hemolytic anemia and liver cell injury, while they are significantly elevated in inflammatory conditions, trauma, and coronary artery disease as part of the acute-phase response.
    Assay Principle

    Haptoglobin present in the sample specifically reacts with anti-human haptoglobin antibodies contained in the reagent to form insoluble immune complexes, resulting in increased turbidity. The degree of turbidity is directly proportional to the haptoglobin concentration in the sample. By measuring the change in absorbance at 340 nm and applying a multipoint calibration curve, the haptoglobin concentration can be accurately quantified.
